Dental

Dental devices and materials are most often evaluated as medical devices, though there are special needs to address. Some dental devices may have exposure to some or all of the following: dentin, mucosal tissue, salivary enzymes or foods. In other cases, the device may be implanted and may be evaluated much like an orthopedic implant device. Typical products in this category include restorative materials (alloys, ceramics, polymers), prosthetic materials, UV curable materials, drug release and other bone or soft tissue implant materials.

Standards and Regulation

Dental devices are evaluated against ISO 10993 for routine biological tests, and according to vertical standards for any specialty tests required (ISO 7405). Animal models can be found in the literature and in reference texts (e.g. Stanley, H., Toxicity Testing of Dental Materials).

Test Categories

The following tests are considered for dental materials and devices:

Characterization of Materials

  • Thorough chemical characterization commensurate with clinical exposure should be completed. The particularly harsh environment in the mouth dictates complete characterization of materials as well as potential extracted chemicals. Simulated degradation studies may also be appropriate.

R & D Function and Performance

  • Test methods that simulate clinical exposure
  • Procedures to satisfy ISO, FDA or ADA requirements

Complete Sterility Assurance Program

  • Sterilization validation
  • Environmental monitoring
  • Packaging validation

Biocompatibility

  • Cytotoxicity of both the material and extracts
  • Acute Toxicity by oral or injectable routes, with and without extracts
  • Acute irritation that may be intracutaneous injection or mucosal tissue contact
  • Delayed contact sensitization

Additional tests that may be required:

  • Implant procedures in muscle or subcutaneous tissue at various intervals
  • Implant procedures in dentin or bone
  • Repeated application to cheek or cheek pouch mucosa
